En el estudio STRIDE-PD, las personas con EP inicial tratadas con LD/entacapona (LCE) presentaron un riesgo mayor de desarrollar discinesias tempranas36 (NE-II).
Un estudio aleatorizado, doble ciego, evidenció que los pacientes no fluctuantes también se beneficiaban del tratamiento con LCE, evidenciando mejores puntuaciones de las actividades de la vida diaria (AVD) y un leve menor requerimiento de LD, con buena tolerancia al tratamiento37 (NE-I).
En un metaanálisis reciente en pacientes con EP inicial que recibían LCE frente a LD, la LCE presentó mejores resultados en cuanto a función motora y actividades de la vida diaria, si bien también se asoció con mayor riesgo de efectos adversos y de abandono38 (NE-I). El estudio EPSILON, aún en curso, pretende evaluar la opicapona en dosis de 50 mg como tratamiento adyuvante a LD en la EP inicial. Los resultados preliminares indican que añadir opicapona a LD supondría una mejoría en función motora y menor riesgo de FC39-41 (NE-I)
